No. The B8 A4 immobilizer does not have a self-programming sequence you can run from the driver's seat. No ignition turn pattern, no button combination. Registering a new fob to this vehicle requires diagnostic equipment connected to the OBD port to run the immobilizer enrollment procedure. The fob you bought online may be the correct one for the vehicle, and the blade can be cut to match. But pairing it to the car is a tools-required job.

Q: One button on my fob stopped working but the others are fine. Do I need a whole new key?

Not necessarily. On the flip-style fob the 2009 A4 uses, individual button contacts on the circuit board can wear out or get contaminated, especially the lock and unlock buttons that take the most use. Sometimes it is residue under the rubber button pad, sometimes the contact point itself is worn. If the fob is otherwise intact and the car still starts normally, the transponder and the remote circuitry are both alive.
