Yes. Once the new remote is programmed to your 2006 Yukon, it communicates with the vehicle's security and body control systems the same way the original did. Whether it works depends on a few things: the procedure has to be done in the correct sequence with specific timing, you need at least one currently working programmed key to initiate it, and the replacement fob has to be a compatible unit to begin with.
